FontUp® and Down Syndrome

A supplement that improves cognitive ability in people with Down syndrome

In recent years, scientific research has studied various natural molecules that can interact with processes related to brain development and cognitive function.

One of the most researched molecules in this context is epigallocatechin gallate (EGCG), a polyphenol found in green tea.

Several scientific teams have analyzed the role of EGCG in relation to certain proteins involved in neuronal development, which has led to preclinical studies and clinical trials in people with Down syndrome.

The formulation developed to facilitate clinical research on this molecule is FontUp®, a dietary supplement designed to provide EGCG with standardized dosing.

What is FontUp®

FontUp® is a food supplement formulated to provide epigallocatechin gallate (EGCG) in capsules with a standardized dose.

How to calculate the correct dosage

In clinical trials conducted with EGCG in Down syndrome, the dosage is adjusted based on body weight. The guideline used in research has been: 10 mg of EGCG per kilogram of body weight per day.

This strategy allows adapting the administered amount to the individual characteristics of each person. Indicative dosage examples:

WEIGHT
APPROXIMATE DOSE
CAPSULES
10 kg 100 mg EGCG 1 capsule
20 kg 200 mg EGCG 2 capsules
66.14 lbs 300 mg EGCG 3 capsules
40 kg 400 mg EGCG 4 capsules
50 kg or more 500 mg EGCG 5 capsules
The maximum dose is 5 capsules per day and it is recommended to take them with meals following a 2-1-2 pattern.

SCIENTIFIC EVIDENCE

DYRK1A: A Key Protein in Down Syndrome

One of the main areas of research in Down syndrome focuses on the protein DYRK1A. This gene is located on chromosome 21, so in people with trisomy 21 there is an overexpression of the DYRK1A protein.

DYRK1A participates in various biological processes related to brain function, including:

Neural development
Formation of neural connections
Brain plasticity

Various preclinical studies have suggested that EGCG can modulate the activity of this protein, which generated interest in evaluating its possible impact on cognitive function.

The TESDAD trial: EGCG in adults with Down syndrome

One of the most well-known clinical studies in this field is the TESDAD trial. This study was a phase 2, randomized, double-blind, placebo-controlled trial, conducted in young adults with Down syndrome.

The study's objective was to evaluate the effect of FontUp® combined with structured cognitive training over a prolonged intervention period.

The study results were published in the scientific journal The Lancet Neurology, one of the most relevant international publications in neurology.

The trial observed improvements in certain cognitive and adaptive domains in participants who received the combination of FontUp® and cognitive training, providing initial evidence of the possible interaction between molecular interventions and cognitive stimulation.

Furthermore, the treatment showed a favorable safety profile during the study period.

The PERSEUS study: EGCG safety in children

Subsequently, the PERSEUS study was developed, designed to evaluate the safety and tolerability of FontUp® in pediatric patients with Down syndrome.

This trial was a phase Ib, randomized, placebo-controlled study that included children between 6 and 12 years of age. Participants received FontUp® or placebo for six months of treatment, followed by an additional follow-up period.

The study results were published in the scientific journal Genetics in Medicine, one of the leading international publications in medical genetics.

The trial showed that the intervention was safe and well tolerated, with no increase in serious adverse events in the treated group.

FontUp® can be part of this global approach as a supplement within programs that promote cognitive health care and general well-being.

Whenever the incorporation of food supplements is considered, it is advisable to do so individually and with the advice of healthcare professionals.

Our commitment is to continue sharing rigorous scientific information and to support families and professionals who work every day to promote the well-being and quality of life of people with Down syndrome.

Frequently Asked Questions

What is FontUp®?

FontUp® is a food supplement containing EGCG (epigallocatechin gallate), a polyphenol purified from green tea.

Each capsule provides 100 mg of EGCG in a standardized formulation.

Why has EGCG been studied in Down syndrome?

In Down syndrome, there is an overexpression of the DYRK1A gene, located on chromosome 21. This protein is involved in various processes related to brain development.

Several preclinical studies have suggested that EGCG can modulate the activity of this protein, which has motivated clinical studies to evaluate its safety and its possible role in cognitive function.

Can the capsules be opened?

Yes. FontUp® capsules can be opened and their contents mixed with food or beverages if the person has difficulty swallowing them.

What is the recommended dose?

The dose used in clinical studies is adjusted to body weight: 10 mg of EGCG per kilogram of body weight per day.

Each capsule of FontUp® contains 100 mg of EGCG.

· For 10 kg of weight -> 1 capsule (100mg EGCG)

· For 20 kg of weight -> 2 capsules (200mg EGCG)

· For 30 kg of weight -> 3 capsules (300mg EGCG)

· For 40 kg of weight -> 4 capsules (400mg EGCG)

· For 50 kg of weight or more -> 5 capsules (500mg EGCG)

The maximum dose is 5 capsules per day and it is recommended to take them with meals in a 2-1-2 regimen.

From what age can FontUp® be taken?

In clinical studies conducted with EGCG in Down syndrome, both young adults and school-aged children have been included.

In any case, the dose should always be adjusted to body weight, and for children, it is advisable to evaluate its use with the pediatrician or healthcare professional who is monitoring them.

Does FontUp® contain gluten or caffeine?

No. FontUp® does not contain gluten or caffeine.

Do you need a prescription?

No. FontUp® is a food supplement and does not require a medical prescription.

Can it be taken with other medications?

No interactions between FontUp® and other medications have been reported in clinical studies. However, if the person is taking medication or has any medical condition, it is advisable to consult their healthcare professional beforehand.

How long can FontUp® be taken for?

In clinical studies with FontUp®, administration periods have been several months and even a year.

As with other food supplements, FontUp® can be used continuously for extended periods, always adapting the duration and dose to individual needs and, especially in the case of children or people taking medication, with the guidance of a healthcare professional.

Is FontUp® a treatment for Down syndrome?

No. FontUp® is a food supplement and is not indicated as a treatment for Down syndrome. Scientific research has studied EGCG in this context, but its use should always be understood as part of a comprehensive approach to care and support.
